In order to analyse astigmatism 3D localization microscopy experiment a calibration for the shape change of the PSF depending on the axial positions is needed. LookUpSTORM only supports the fitting of single emitter signals by using a lookup table. However, for a calibration fitting methods with free parameters (for the covariance) are needed. Therefore, LookUpSTORM supports the usage of calibation files (YAML) created from ThunderSTORM. In order to run the calibration, download and install the ThunderSTORM plugin and select Plugins -> ThunderSTORM -> 3D calibration -> Cylindrical lens calibration in the ImageJ menu.
Below an example of creating a calibration file using ThunderSTORM from beads (using the SMLMS challange dataset) can be seen.
